Description:AGO2/Argonaute-2 Protein is vital for RNA-mediated gene silencing through the RNA-induced silencing complex (RISC). 'Minimal RISC' involves AGO2 bound to guide RNA, such as microRNA (miRNA) or short interfering RNA (siRNA), guiding RISC to complementary mRNAs for silencing. The mechanism depends on complementarity; perfect matches lead to mRNA cleavage, while partial matches inhibit translation. AGO2 may hinder translation initiation by binding to the cap or interact with EIF6 to inhibit ribosomal subunit association. It also participates in up-regulating translation during specific conditions, such as serum starvation, by binding to the AU element in the 3'-UTR of TNF mRNA. AGO2 is crucial for transcriptional gene silencing (TGS) and regulates lymphoid and erythroid development independently of its endonuclease activity.
